Design Notes

We designed three hybrid promoters combining Plac or PlacUV5, lacO and lldRO1 and lldRO2. Part:BBa_K1847010, Part:BBa_K1847011 and Part:BBa_K1847012. In Part:BBa_K1847010 and Part:BBa_K1847011 we kept the original architecture of the PlldR leaving the operators and substituting the wild-type promoter by Plac and lacO. The distance in nucleotides between the two lldR operator sites differs in 5 nucleotides. In Part:BBa_K1847012 the lldR wild-type promoter was substituted by a biologically neutral spacer and Plac and lacO were located in front of the PlldR.
